嵌入分子诱导的DNA熵弹性转变

摘要
我们使用光镊对DNA分子与可嵌入DNA分子的药物柔红霉素和溴化乙锭相互作用时进行拉伸实验。这些实验在从零到2 pN的低力区间内进行。我们的结果表明,随着药物浓度增加至某一临界值,DNA-药物复合物的持久长度强烈增加。超过该临界值后,持久长度急剧下降,并在更高药物浓度下基本保持恒定。分子的围长随药物浓度增加而单调增加并趋于饱和。测量得到的嵌入剂临界浓度与先前由沉降实验获得的DNA-药物复合物螺旋-线团转变的报道值一致。
