利用兆赫兹X射线自由电子激光脉冲解析抗体蛋白的分子扩散与聚集

摘要
兆赫兹重复频率的X射线自由电子激光(XFELs)可为生物大分子溶液的结构动力学提供新颖见解。然而,极高的剂量率会因辐射损伤引发束流诱导的动力学与结构变化。在此,我们利用欧洲XFEL的兆赫兹X射线光子关联谱(MHz-XPCS)探测致密抗体蛋白(Ig-PEG)溶液的动力学。通过改变总剂量与剂量率,我们确定了测量蛋白质在其第一配位层内运动的区间,量化了如驱动运动等XFEL诱导效应,并描绘了团聚动力学的范围。结果表明,在平均剂量率低于 、时间窗口达 时,可在束流诱导聚集发生前捕获蛋白质动力学。我们将此方法称为聚集前关联,并证明MHz-XPCS弥补了生物样品测量技术中重要的时空空白。
