预转变效应介导跨膜蛋白间的组装力

摘要
我们提出了一种脂质双分子层中跨膜蛋白通用且强大的组装与迁移力机制。该力是宿主膜中有序相与无序相之间一级相变的预转变(或预熔融)效应。利用大规模分子模拟,我们表明具有与无序相相等疏水厚度的蛋白质嵌入有序双分子层中时,会稳定一个微观的有序-无序界面,且该界面的刚度有限。当两个这样的蛋白质相互接近时,它们发生组装,因为组装降低了净界面自由能。类比疏水效应,我们将此现象称为“orderphobic effect”。该效应由接近有序-无序相变以及蛋白质的尺寸和疏水失配所介导。源于 orderphobic effect 的力强度和作用范围远大于我们所考察膜中由膜弹性产生的力。
