激光扫描显微镜中驻波激发的多平面荧光揭示红细胞三维信息

摘要

荧光驻波激发在光学显微镜中备受推崇,因为它能提高轴向分辨率。我们在此证明,通过在样品附近放置平面反射镜,可在单点激光扫描显微镜中产生由驻波激发的多平面荧光。我们报告称,每个激发平面内的相对强度取决于荧光团的斯托克斯位移。我们通过使用针对细胞膜的特异性染料,展示了如何利用驻波激发生成红细胞表面膜的精确等高线图,其轴向分辨率约为 90 nm。该方法仅需在现有的共聚焦激光扫描显微镜中添加一面平面镜,可能在研究涉及红细胞膜的疾病(如疟疾)方面证明是有用的。
