橄榄狒狒(Papio anubis)感染诺氏疟原虫 H 株后的妊娠疟疾免疫应答

摘要

撒哈拉以南非洲约有 2400 万孕妇面临罹患妊娠疟疾并发症的风险。导致孕妇对疟疾易感性增加的机制尚未完全阐明。狒狒对诺氏疟原虫(Plasmodium knowlesi)易感,且其生殖生理及宿主-病原体相互作用与人类相似,这使其有望成为研究妊娠疟疾机制的理想模型。本研究利用狒狒对诺氏疟原虫感染的易感性,表征了感染诺氏疟原虫的狒狒妊娠疟疾病理发生过程中涉及的细胞因子和外周血单个核细胞回忆增殖应答。通过超声检查确认了三只定时交配的成年雌性狒狒的妊娠状态及孕龄。在妊娠第 150 天,使这三只怀孕狒狒及四只未孕对照狒狒感染诺氏疟原虫 H 株寄生虫。随后每周采集外周血清及单个核细胞。采用酶联免疫吸附试验(ELISA),以相应的酶标抗体检测血清细胞因子浓度。同时每周进行外周血单个核细胞回忆增殖试验。结果表明,该模型中的妊娠疟疾与干扰素 γ 及白细胞介素 6(IL-6)应答的抑制相关。肿瘤坏死因子 α 应答上调,而 IL-4、IL-12 及回忆增殖应答与对照组无差异。这些数据在很大程度上与人类研究中的一些发现一致,表明该模型用于研究妊娠疟疾相关机制的可行性。
