用于细胞膜成像的共焦超临界角荧光显微镜

摘要

我们展示了利用传统共焦显微镜对生物样品进行亚波长层析成像。这种光学层析是通过超临界角荧光现象实现的,其中只有位于折射率不连续界面附近的荧光团才能将辐射的传播分量发射到所谓的“禁戒角”中。该技术的简便性使其仅需对探测通道进行简单修改,即可集成到高数值孔径共焦扫描显微镜中。共焦 -SAF 显微镜将成为实现高分辨率表面成像的有力工具,尤其适用于生物样品的膜成像。
