共焦显微镜中基于正弦相位合成光学全息术的可获取定量相位成像

摘要
我们提出了一种基于正弦相位参考波合成光学全息术的共焦显微镜定量相位成像的技术简便实现。利用Mirau干涉物镜与压电控制载物台的低幅垂直样品振动,我们在商用共焦显微镜(尼康,型号:A1R;蔡司,型号:LSM-880)上记录合成全息图,并从中重建定量相位图像。我们通过无染色成像宫颈(HeLa)与卵巢(ES-2)癌细胞以及干细胞(mHAT9a)样本展示了我们的技术。我们的技术有望通过提供无标记细胞寻找、监测细胞形态以及基于定量相位衬度的活细胞非扰动长时间观察,来拓展共焦显微镜中的荧光成像应用。
