利用 X 射线激光对纳米颗粒系综进行三维衍射成像

摘要

我们报道了利用 X 射线单颗粒成像(SPI)测定金纳米颗粒(AuNPs)三维结构的结果。在欧洲 X 射线自由电子激光装置的新能力支持下(可每秒测量 1500 帧),在不足 100 小时的束流时间内测量了约 1000 万张金纳米颗粒衍射图样,数据量超过此前任何单次 SPI 实验的 100 倍。我们开发了分类与结构排序方法以厘清颗粒的非均匀性,并获得优于 3 nm 的分辨率。借助这些新的实验与分析进展,SPI 方法已进入新时代,且迈向近原子分辨率生物分子成像的路径已清晰可见。
