无扫描的宽场双光子激发：高时间分辨与低光漂白的活细胞显微术

摘要
我们展示了如Hwang及其合作者先前所述但在更大视场和高达100 Hz帧率下生物样品中的无扫描双光子激发荧光成像。在记录负载荧光Ca指示剂Fluo-4 AM的原代大鼠神经元培养中突触驱动的Ca事件时，我们观察到与单光子激发相比光漂白大幅降低。该方法无需对显微镜作昂贵添置，有望用于对高时间分辨率有需求的工作。
