斑马鱼因其高繁殖率和快速生长速度,成为研究化学、基因及环境扰动对发育影响的理想模型。最近,单细胞测序方法日益成为衡量这些扰动对整个胚胎尺度影响的强大工具。此类实验依赖于能够并行从大量 individually 标记的斑马鱼胚胎中分离核糠体。本文报告了一种通过珠子脱落于破裂缓冲液中,以高效分离斑马鱼胚胎中高质量核糠体的方法。通过 head-to-head sciPlex-RNA-seq 实验,我们展示了该方法在 enzymatic 脱 dissociation 方面具有显著优势,且适用于广泛的发育阶段。
